On Thursday, Aptose Biosciences Inc. (NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/APTO#NASDAQ">APTO</a>) announced that the Cohort Safety Review Committee monitoring Phase 1/2 TUSCANY trial of tuspetinib in combination with standard of care dosing of venetoclax and azacitidine (TUS+VEN+AZA triplet) has approved escalating from 40 mg TUS to 80 mg TUS based on its favorable review of data from the first <a target="_blank" href="https://www.benzinga.com/pressreleases/25/02/g43844887/aptose-announces-positive-clinical-safety-review-committee-csrc-approval-to-dose-escalate-in-phase" rel="noreferrer noopener">four patients in the trial</a>.
The TUS+VEN+AZA triplet is being developed as a frontline therapy to treat large, mutationally diverse populations of newly diagnosed acute myeloid leukemia (AML) patients who are ineligible to receive induction chemotherapy.
No significant safety concerns or dose-limiting toxicities (DLTs) have been reported, including no prolonged myelosuppression of subjects in remission. All four subjects treated in the 40 mg cohort remain in the study, while enrollment is open for the 80 mg cohort.
Multiple U.S. sites are enrolling in the TUSCANY trial, with an anticipated enrollment of 18-24 patients by mid-late 2025. Data will be released as it becomes available.
Three patients with unmutated (wildtype) FLT3 (FLT3-WT) completed Cycle 1 of treatment with no dose-limiting toxicities (DLTs) and no TUS dose adjustments. Two FLT3-WT patients achieved complete remissions (CR and CRh) by the end of Cycle 1. A patient with biallelic TP53 mutations and a complex karyotype obtained CR.
The third FLT3-WT patient experienced significant reductions in bone marrow leukemic blasts during Cycle 1 and remains on therapy in Cycle 2.
The fourth patient, harboring FLT3-ITD and NPM1 mutations, is currently dosing in Cycle 1 and is not yet eligible for response evaluation.
Pharmacokinetic (PK) analyses for TUS show plasma levels unaffected by adding AZA.
Price Action: APTO stock is up 31.50% at $0.20 at last check Thursday.

U.S. stock futures fell on Wednesday after scaling fresh records in Tuesday&#8217;s session. The futures of all four benchmark indices were trading lower in the premarket.
President Donald Trump, on Tuesday, announced plans to impose a 25% tariff on auto imports, along with tariffs on pharmaceuticals and semiconductor chips.
The auto tariffs are expected to take effect by April 2, based on a report from his cabinet. Tariffs on pharmaceuticals and chips will also start at 25% or higher, with increases over the year. Trump did not specify a start date, allowing manufacturers time to establish U.S. factories to avoid the tariffs.
The 10-year Treasury yield stood at 4.56%, while the two-year yield was at 4.30%. According to the CME Group's FedWatch <a href="https://www.cmegroup.com/markets/interest-rates/cme-fedwatch-tool.html">tool</a>, there is a 97.5% chance that the Federal Reserve will keep interest rates unchanged for the March meeting.

Despite the overall positive trend, communication services and consumer discretionary sectors declined on Tuesday. 
The broader market awaited the release of the latest FOMC minutes on Wednesday and monitored other ongoing geopolitical developments.
Last week saw positive momentum across equity markets. The S&amp;P 500 was approximately up 1.5%, the Dow Jones Industrial Average rose 0.6%, and the Nasdaq Composite gained 2.6% last week.
In company news, Fluor Corp. (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/FLR#NYSE">FLR</a>) shares plummeted over 8% after the company reported quarterly earnings and revenue below expectations. On the economic front, the NY Empire State Manufacturing Index exceeded forecasts, rising 18.3 points to +5.7 in February.

Insights From Analysts
Ryan Detrick, the chief market strategist at Carson Research, pointed out that the S&amp;P 500 scaled its second all-time high of 2025 on Tuesday.
This was the 1,243rd record high over the 17,176 trading days since 1957 when the gauge increased to having 500 stocks.

Talking about the fresh record, Nathan Peterson, director of derivatives analysis at the <a href="https://www.schwab.com/learn/story/stock-market-update-open">Schwab Center</a> for Financial Research said: &#8220;It seems that stocks can maintain a bullish bias provided the economic data and earnings growth estimates hold up and yields on the 10-year Treasury note remain below the mid-January high of 4.8%." 
According to economist Jeremy Siegel, despite the uncertainties, "The stock market continues to perform well, particularly in the tech sector, even as leadership has broadened beyond last year's AI-driven rally."
Professor of finance at the University of Pennsylvania, Siegel also added that "Tesla took a hit on competition from China's BYD, now the world's largest EV maker. Nvidia was hit after the DeepSeek news, but overall, the tech leaders have remained resilient."

Cues From The Last Session
Tech and Communication Services bucked the negative trend, closing higher on Friday, while Healthcare, Consumer Staples, and Utilities led losses as most S&amp;P 500 sectors declined. 
Despite Friday&#8217;s mixed results, for the last week, the S&amp;P 500 gained 1.5%, the Dow 0.6%, and the Nasdaq 2.6% led by weaker-than-expected retail sales which were down 0.9% in January.

Ryan Detrick, the chief market strategist at Carson Research pointed out that seven out of the total broad 11 sectors have outperformed the S&amp;P 500 index on a year-to-date basis.

The S&amp;P 500 has surged 60% in the past two years, a significant increase. However, according to Bravos Research historically, similar rapid gains have often been followed by substantial market downturns.
The downturns have occurred three out of the last four times after such a surge was witnessed. Bravos highlights that one instance did lead to a rapid and dramatic market rise, calling it a &#8220;melt-up.&#8221;
The question, now is whether the current situation will follow the more common pattern of decline or the rarer &#8220;melt-up&#8221; scenario.

Talking about the economic impact Ed Yardeni in his Market Call blog said, &#8220;So far, Trump&#8217;s tariff turmoil seems to be weighing more on the U.S., Canada, Mexico, and many other emerging markets than on China and most European countries.&#8221;
That could change once reciprocal tariffs are imposed by the U.S. in early April, unless Trump changes his mind, again, added Yardeni.

Shares of FedEx Corporation (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/FDX#NYSE">FDX</a>) rose sharply in today's pre-market trading as the company posted stronger-than-expected <a href="https://www.benzinga.com/news/earnings/24/12/42605034/fedex-stock-rallies-on-better-than-expected-q2-eps-freight-spin-off-announcement">results for its second quarter</a>.
The company also announced that it has decided to pursue a full separation of FedEx Freight through the capital markets, creating a new publicly traded company.
FedEx shares jumped 8.7% to $299.80 in the pre-market trading session.
