Roche (OTC: <a class="ticker" href="https://www.benzinga.com/stock/rhhby#OTC">RHHBY</a>) announced that the European Commission has granted marketing authorization to the subcutaneous&nbsp;formulation of relapsing multiple sclerosis (RMS) and primary progressive multiple sclerosis (PPMS) drug Ocrevus (ocrelizumab).
The approval is based on pivotal data from the late-stage OCARINA II trial, which showed non-inferior levels of the drug in the blood when administered subcutaneously, and a safety and efficacy profile comparable to the intravenous (IV) formulation in patients with RMS and PPMS.
The SC formulation of Ocrevus was developed to provide an alternative twice-a-year treatment option to patients and healthcare professionals, in addition to IV.
Ocrevus SC combines Ocrevus with Halozyme Therapeutics&#39; ENHANZE drug delivery technology.
Roche also announced that the European Medicines Agency (&quot;EMA&quot;) has validated and initiated review of the marketing authorization application&nbsp;for Elevidys (delandistrogene moxeparvovec).Elevidys, a gene therapy for the treatment of ambulatory patients aged 3-7 years with Duchenne muscular dystrophy, is already approved in the United States and some other countries. It is a one-time treatment administered through a single IV dose.
The MAA is based on results from the phase III EMBARK study, a global, randomized, double-blind, placebo-controlled study in DMD patients aged 4 through 7 years. Although the EMBARK study did not meet the primary endpoint, the totality of evidence from the trial confirmed that Elevidys is the first gene therapy to provide clinically meaningful benefits for patients by modifying the disease course, with a manageable safety profile.
Elevidys-treated patients reported clinically meaningful and statistically significant benefits in both key secondary functional endpoints &mdash; time to rise from the floor and a ten-minute walk/run test.
In addition, a clinically meaningful and statistically significant improvement was observed for the pre-specified secondary endpoint &mdash; stride velocity 95th centile.
The MAA is also supported by data from another study in DMD patients, ENDEAVOR, an open-label study that is enrolling ambulatory and non-ambulatory patients of various ages, along with a phase I/II study which provides long-term efficacy, durability, safety and biological data to support the benefit-risk assessment.
Per Roche, a potential approval will make Elevidys the first and only gene therapy to address the underlying cause of DMD, available in Europe.
Two other studies are also currently ongoing to expand the drug&#39;s label ENVOL, a phase II study in boys with DMD under the age of 4 and ENVISION, a phase III study in older ambulatory and non-ambulatory patients with DMD.
Roche has a collaboration agreement with Sarepta Therapeutics (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/srpt#NASDAQ">SRPT</a>) for Elevidys, whereby it is responsible to commercialize Elevidys outside the United States.
Last week, Sarepta announced that the FDA has converted the accelerated approval granted to Elevidys to a traditional approval and expanded the label to include ambulatory individuals with a confirmed mutation in the DMD gene who are aged four and older. In addition, the FDA also granted accelerated approval to Elevidys for non-ambulatory Duchenne patients.
Roche&#39;s shares have lost 1.4% year to date against the industry&#39;s growth of 22.1%.

Ultragenyx Pharmaceutical Inc. (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/rare#NASDAQ">RARE</a>), along with partner Mereo BioPharma (MREO), announced new positive 14-month data from the phase II portion of the phase II/III Orbit study evaluating setrusumab (UX143) in patients with osteogenesis imperfecta.
The data showed that the large reduction in fracture rates previously reported in patients treated for a minimum of six months was sustained in patients treated for at least 14 months.
OI is a group of genetic disorders that impact bone metabolism. It is characterized by increased bone fragility and a high risk of fractures. Currently, there are no approved treatments for this debilitating condition, representing a highly unmet medical need.
As of the data cut-off date of May 24, 2024, the new data showed that treatment with setrusumab resulted in&nbsp;a large, sustained 67% reduction in annualized fracture rate and persistent median annualized fracture rate of 0.00.
Treatment with setrusumab also resulted in clinically meaningful continued improvement in lumbar spine bone mineral density (BMD) with a mean increase from baseline of 22% at the 12-month timepoint.
Previously, the company reported that treatment with setrusumab for at least six months reduced the annualized fracture rate by 67% in patients with OI.
Per the company, the new data underlines the potential of setrusumab in reducing the incidence of fractures and improving BMD in patients with OI.
No treatment-related serious adverse effects were seen as of the data cut-off date.
Shares of Ultragenyx have plunged 13.4% so far this year compared with the industry&#39;s decline of 5.5%.

Setrusumab, a fully human monoclonal antibody, is an inhibitor of the sclerostin protein, which is responsible for hampering the maturation and activity of bone-forming cells.
We would like to remind investors that Ultragenyx entered into a licensing and collaboration agreement with Mereo in December 2020 to co-develop setrusumab. Per the terms of the agreement, Ultragenyx leads the global development of setrusumab in both pediatric and adult patients with OI.
As part of this deal, Mereo received an upfront payment of $50 million from RARE, while the latter is eligible for certain regulatory and sales-based milestone payments.
Meanwhile, enrollment in the pivotal phase III portion of the phase II/III Orbit study has been completed. Patients in this portion of the study will be divided into two cohorts in a ratio of 2:1, receiving either a 20 mg/kg dose of setrusumab or a placebo. The primary efficacy endpoint of the study is the annualized clinical fracture rate. Per Ultragenyx, all patients will transition to an extension period and receive open-label setrusumab after completing the primary analysis in the phase III portion of the study.
Apart from the Orbit study, the phase III Cosmic study is also evaluating the effect of setrusumab compared with intravenous bisphosphonate therapy on the annualized total fracture rate in patients aged 2 to &lt;7 years. Enrollment in this study has also been completed.

Candel Therapeutics, Inc. (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/cadl#NASDAQ">CADL</a>) announced overall survival data from a phase II study evaluating its lead investigational adenovirus immunotherapy candidate, CAN-2409 plus valacyclovir (prodrug), together with standard of care (SoC) in patients with stage III/IV non-small cell lung cancer (NSCLC) who are non-responsive to immune checkpoint inhibitor therapy.
Data from the study showed that treatment with two administrations of CAN-2409 plus valacyclovir led to a median overall survival of 20.6 months in NSCLC patients whose disease had progressed despite receiving prior anti-PD-1 treatment.
The data will be presented at the annual meeting of the American Society of Clinical Oncology, which is scheduled to be held from May 31 to Jun 4, 2024.
Per the press release, the above mentioned data was in context to earlier published data, which demonstrated a median overall survival of 11.6 months observed with standard-of-care docetaxel-based chemotherapy in a similar patient population.
Importantly, treatment with CAN-2409 exhibited a favorable safety and tolerability profile in NSCLC patients as of the data cut-off date.
Despite the upbeat data, shares of Candel were down in pre-market trading on May 23. The stock has skyrocketed 627.2% year to date against the industry&#39;s decline of 4.3%.

<li>&nbsp;The NDA submission lacks the data needed to establish that the change in negative symptoms of schizophrenia with roluperidone treatment was clinically meaningful.</li>
<li>&nbsp;The submitted safety database included an inadequate number of subjects exposed to roluperidone at the proposed dose (64 mg) for at least 12 months.</li>
<li>&nbsp;To address these deficiencies, the FDA stated that Minerva must submit at least one additional positive, adequate, and well-controlled study to support the safety and effectiveness of roluperidone for treating negative symptoms.&nbsp;</li>
<li>&nbsp;Minerva must also provide additional data to demonstrate the safety and efficacy of roluperidone co-administered with antipsychotic medications, to support that observed effect on negative symptoms with roluperidone treatment corresponds to a clinically meaningful change, and to demonstrate the long-term safety of the proposed dose.</li>
</ul>
&#8220;We believe that roluperidone is a safe and effective therapy for negative symptoms of schizophrenia, and we will review FDA&#8217;s feedback and consider our potential paths forward, including continuing to work closely with the FDA and providing any additional information as needed,&#8221; said Dr. Remy Luthringer, Executive Chairman and CEO of Minerva.
In addition to the clinical deficiencies, the FDA also provided comments on, among other items, clinical pharmacology, product quality, biopharmaceutics, and nonclinical issues.

Bank lending to households in the Eurozone rose by 0.3% year-over-year to EUR 6.870 trillion for January. The consumer confidence indicator in France declined to 89 in February versus around two-year high level of 91 in the prior month. The GfK Consumer Climate Indicator for Germany climbed to a reading of -29.0 heading into March compared to February&#39;s level of 29.6.

&nbsp;
Asian markets closed higher on Tuesday, with Japan&rsquo;s Nikkei 225 gaining 0.01%, Hong Kong&rsquo;s Hang Seng Index climbing 0.94%, China&rsquo;s Shanghai Composite Index surging 1.29% and India&rsquo;s S&amp;P BSE Sensex gaining 0.4%.
Hong Kong recorded a trade surplus of $3.6 billion in January compared to a year-ago deficit of $25.4 billion. The annual inflation rate in Japan slowed to 2.2% in January compared to 2.6% in the earlier month.

U.S. durable goods orders fell by 6.1% month-over-month in January compared to a 0.3% decline in December.
The FHFA house price index rose 0.1% in December following a 0.4% increase in November.
The S&amp;P CoreLogic Case-Shiller 20-city home price index climbed by 6.1% year-over-year in December.
The Dallas Fed general business activity index for Texas&rsquo; service sector rose to -3.9 in February from -9.3 in the previous month.

Crude Oil Gains 1%; Macy's Plans To Close 150 Stores

U.S. stocks traded mixed this morning, with the Nasdaq Composite gaining around 50 points on Tuesday.
Following the market opening Tuesday, the Dow traded down 0.07% to 39,043.13 while the NASDAQ rose 0.31% to 16,026.43. The S&amp;P 500 also rose, gaining, 0.10% to 5,074.75.

Lowe&rsquo;s reported fourth-quarter FY23 sales of $18.60 billion, beating the analyst consensus estimate of $18.45 billion. EPS of $1.77 beat the consensus estimate of $1.68.
For FY24, Lowe&rsquo;s expects revenue of $84 billion-$85 billion (against the Street view of $85.61 billion) and adjusted EPS of $12.00 &ndash; $12.30 vs. consensus of $12.95. The company expects comparable sales to decline about 2% to 3% in FY24.

Janux Therapeutics, Inc. (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/janx#NASDAQ">JANX</a>) shares shot up 138% to $36.00 after the company announced updated clinical data for both of its clinical programs.
Shares of Infobird Co., Ltd (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/ifbd#NASDAQ">IFBD</a>) got a boost, surging 111% to $5.84. The stock continues to experience marked volatility after the company on Monday said it encountered an unexpected delay in its planned reverse stock split.
Viking Therapeutics, Inc. (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/vktx#NASDAQ">VKTX</a>) shares were also up, gaining 79% to $68.93 after the company announced the Phase 2 VENTURE trial successfully achieved its primary endpoint and all secondary endpoints.
