Dubbed JN.1, the omicron variant is highly contagious and accounts for <a href="https://www.reuters.com/business/healthcare-pharmaceuticals/cdc-says-jn1-variant-accounts-about-86-covid-cases-us-2024-01-22/">more than 85% of the reported cases</a> nationwide. It is now the <a href="https://publichealth.jhu.edu/2024/jn1-the-dominant-variant-in-the-covid-surge">dominant strain in the U.S.</a> and around the world and according to the U.S. Centers for Disease Control and Prevention, and it could be &ldquo;intensifying&rdquo; the spread of COVID-19. At the same time JN.1 is spreading, <a href="https://www.cdc.gov/coronavirus/2019-ncov/science/forecasting/forecasting-hospitalizations-previous-2024.html">COVID hospitalizations</a> are rising, prompting some healthcare facilities to require masks again.&nbsp;
The new variant is also responsible for what is already called the second-largest surge, following the omicron breakout in late 2021 and early 2022. That surge infected more people than at the start of the pandemic in March of 2020. Meanwhile, long COVID-19 is still largely a mystery to doctors and scientists who have seen patients suffer from fatigue and other symptoms that last months after the illness is over.&nbsp;
Many existing viruses continue to make circuits of epidemics around the world.&nbsp;
<h2>Disease X On The Horizon&nbsp;</h2>
If that isn&rsquo;t bad enough, scientists are warning of another global pandemic that could be 20 times worse than COVID-19. Called <a href="https://www.who.int/news-room/events/detail/2024/01/19/default-calendar/Research-response-to-pathogen-X-during-a-pandemic">Disease X,</a> scientists at the World Health Organization believe the next pandemic could be caused by a new pathogen, not a variant of a current disease. If those predictions prove true, It would have a huge negative impact on healthcare systems and economies around the world since not enough has been done to prepare for future pandemics.
Sure, the World Bank established the Pandemic Fund, which is focused on helping low and middle-income countries strengthen their pandemic preparedness, but it may not be enough. To date, the fund <a href="https://www.worldbank.org/en/news/press-release/2023/12/22/the-pandemic-fund-announces-second-round-of-funding-with-half-a-billion-dollar-envelope">mobilized $2 billion</a> but some estimates call for the need for upwards of <a href="https://www.kff.org/global-health-policy/issue-brief/the-new-pandemic-fund-overview-and-key-issues-for-the-u-s/#:~:text=Donors%20have%20promised%20over%20%241.9,%2C%20WHO%2C%20and%20other%20groups.">$10 billion a year</a> to tackle new outbreaks. The Global Preparedness Monitoring Board (GPMB), an independent body that assesses the world&rsquo;s preparedness for pandemics and other health emergencies, warned that in the wake of COVID-19, the world is woefully unprepared to deal with potential new pandemics.&nbsp;
<h2>Limitations Of Vaccines And Antibodies Are Now Well Known</h2>
The COVID-19 pandemic clearly demonstrated the deficiencies of the current way of responding to the pandemic. Vaccines and antibodies, always considered reliable workhorses, can be readily escaped by viruses and become less and less useful if not completely useless.&nbsp;&nbsp;
Nor is the current approach of developing one or more drugs for each potential viral threat a satisfactory one. Firstly, it is very expensive in terms of the costs of drug development and approval, and later on to keep the stockpiles going, and secondly, it requires that the scientists know the causative agent years ahead of the pandemic.&nbsp;
And none of these approaches would work in responding to a Disease X scenario.&nbsp;
A novel approach is clearly needed. One approach that could work is a drug that works against many viruses of different kinds, so even as the virus changes in the field, it will not escape the drug.&nbsp;
Is that even possible?&nbsp;
Think of antibiotics. Before the discovery of penicillin, there were no drugs that could treat the myriads of bacterial infections that were killing people. And the one penicillin works against many of those bacteria. That led to an explosion of developments to find similar drugs and modified penicillins that provided the ability to take the drug by mouth instead of injection, improve their effectiveness, etc. Amoxicillin, discovered in 1958, is even today the most commonly used oral antibiotic for children, over 65 years later! That is the great thing about such super-broad-spectrum drugs.&nbsp;
So, is any company working on such a dream drug for viruses? Or is it just science fiction?&nbsp;
<h2>Novel Drug To Fight Many Viruses Is In The Works&nbsp;</h2>
It is science fiction until it becomes reality, right? There is one company that reports it has made great strides and is now very close to the goal of bringing that science fiction to everyday reality.&nbsp;
Imagine the same antiviral drug that works against almost all important respiratory viruses, including the cause of COVID-19, other coronaviruses, RSV and possibly many other respiratory diseases (for example, hMPV &ndash; human metapneumovirus &ndash; which is closely related to RSV). There is one company that says it has already made it possible. The company we are talking about is NanoViricides, Inc. (AMEX: NNVC). This <a href="http://www.nanoviricides.com/aboutus.html">clinical-stage drug company</a> is developing nanomedicine drugs to fight viruses including COVID-19, RSV and shingles.
NV-CoV-2, its leading drug candidate, is based on the company&rsquo;s nanoviricide active pharmaceutical ingredient NV-387, which the company says blocks the reinfection cycle of the viral disease. It binds to the virus and fuses with the virus surface, uprooting the glycoproteins required for the virus to bind to the human cell &ndash; rendering it incapable of infecting a cell. While antibodies can only cover the virus, NV-387 is a chemical nanomachine that completely disrupts the virus. NanoViricides has shown in electron microscopic pictures of viruses that their drugs do indeed disrupt the virus particle. The company likens its drug ingredient to antibiotics in that it can be used to treat a variety of viruses.&nbsp;
NanoViricides has already proven in demanding animal studies that NV-387 is highly effective against coronaviruses, RSV and even Smallpox/Mpox! Thus, the NanoViricides platform technology could be poised to revolutionize the fight against viruses just as antibiotics revolutionized the fight against bacterial infections. And the company is already advancing its first drug through clinical trials toward regulatory approval to make this all real.&nbsp;&nbsp;
<h2>No Adverse Effects In Phase 1 Trial&nbsp;</h2>
Towards that end, the company just completed <a href="https://finance.yahoo.com/news/nanoviricides-inc-filed-annual-report-104500182.html">phase 1 human clinical trials</a> in healthy subjects, using two oral formulations: NV-CoV-2 Oral Syrup and NV-CoV-2 Oral gummies in India. Both include its nanoviricide active agent NV-387. Karveer Meditech Pvt. Ltd., the company&rsquo;s licensee and collaborator, is the drug sponsor in India and has the rights to develop and commercialize the drugs in India.&nbsp;
All 36 healthy human subjects have completed the clinical trial. This included single dosing studies, which were completed around the end of November, and now multiple dosing studies, which the company said were completed just recently. No adverse events or serious adverse events were reported during the entire phase 1 trial in healthy subjects, which indicates that both of the formulations are safe in humans. These results are consistent with the company&rsquo;s preclinical safety/toxicology animal studies that indicated the drug NV-387 is very safe.
There are currently very few, if any safe drugs in the antiviral space. Almost all antiviral drugs have limitations due to dose-limiting toxicities or side effects. This includes the remaining approved COVID-19 drugs, namely Paxlovid&reg; by Pfizer (NYSE: <a class="ticker" href="https://www.benzinga.com/stock/pfe#NYSE">PFE</a>) and Remdesivir, Veklury&reg; by Gilead (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/gild#NASDAQ">GILD</a>). As such, the safety demonstrated by NV-387 is noteworthy.
The JN.1 COVID-19 variant rapidly spreading around the world is a wake-up call. This virus isn&rsquo;t going away, and the potential for even more dangerous ones is ever-present. The world isn&rsquo;t prepared, but NanoViricides wants to change that. It is working hard to develop a new class of drugs that can not only fight COVID-19 and other viral illnesses but prevent them from spreading.&nbsp;

The CDC <a href="https://abcnews.go.com/Health/cdc-warns-rsv-cases-rising-young-children-southeastern/story?id=102994557">recently issued a warning</a> that Respiratory Syncytial Virus (RSV), a common and highly contagious lung infection that can cause bronchitis and pneumonia, is on the rise in the Southeastern United States &ndash; and the very young and the elderly are <a href="https://www.cdc.gov/rsv/high-risk/index.html">the most at risk</a> of developing severe incidences of RSV.&nbsp;
Every year, there are <a href="https://www.cdc.gov/rsv/research/index.html#:~:text=Each%20year%20in%20the%20United,younger%20than%205%20years%20old.&amp;text=58%2C000%2D80%2C000%20hospitalizations%20among%20children%20younger%20than%205%20years%20old.&amp;text=60%2C000%2D160%2C000%20hospitalizations%20among%20adults%2065%20years%20and%20older.">over 2 million outpatient (non-hospitalized) cases of RSV</a> in children under 5, and RSV is the leading cause of hospitalizations for young children. There are <a href="https://www.cdc.gov/rsv/research/index.html#:~:text=Each%20year%20in%20the%20United,younger%20than%205%20years%20old.&amp;text=58%2C000%2D80%2C000%20hospitalizations%20among%20children%20younger%20than%205%20years%20old.&amp;text=60%2C000%2D160%2C000%20hospitalizations%20among%20adults%2065%20years%20and%20older.">up to 80,000 hospitalization cases for children under 5</a>, and there are also up to 160,000 cases of patients over 65 getting hospitalized with RSV. Up to 300 children under 5 and 10,000 adults over 65 die of RSV annually. Despite this serious disease, there were no vaccines or good treatment options until recently.
The recently approved vaccines from GSK plc (NYSE: <a class="ticker" href="https://www.benzinga.com/stock/gsk#NYSE">GSK</a>) and Pfizer (NYSE: <a class="ticker" href="https://www.benzinga.com/stock/pfe#NYSE">PFE</a>) also have severe limitations. These vaccines are only for use in patients aged 60 and over. However, these vaccines can<a href="https://www.verywellhealth.com/new-rsv-vaccines-side-effects-7509954"> involve side effects</a>, including atrial fibrillation, Guillain-Barr&eacute; syndrome and another potentially related disorder. A recently approved vaccine for pregnant women to protect the newborn infant upon birth has the risk of preterm births.&nbsp;
In July 2023, the FDA also approved <a href="https://www.sanofi.com/en/media-room/press-releases/2023/2023-07-17-17-00-00-2705911">Beyfortus</a>, an antibody treatment for children delivered as an injection to help prevent the development of RSV. Beyfortus was developed by Sanofi (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/sny#NASDAQ">SNY</a>) and AstraZeneca (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/azn#NASDAQ">AZN</a>). It is expected to be better than a previously approved antibody (palivizumab, trade name Synagis, developed by MedImmune, now marketed by SOBI &ndash; Swedish Orphan Biovitrum) for the same indication. Like the vaccines, however, there are side effects associated with these protective antibody treatments, which can include <a href="https://reference.medscape.com/drug/beyfortus-nirsevimab-4000354">anaphylaxis</a>, a severe allergic reaction.&nbsp;
Yet, there are no treatments for RSV infection except for severe hospitalized cases when children at risk of dying are given the toxic drug ribavirin at very high doses as a last resort.&nbsp;

The market for therapeutics for RSV was worth <a href="https://www.globenewswire.com/en/news-release/2023/06/21/2691766/0/en/Respiratory-Syncytial-Virus-RSV-Therapeutics-Market-to-Hit-US-8-73-Billion-by-2031-Exclusive-Report-by-Growth-Plus-Reports.html">$1.8 billion in 2022</a> and will reach $8.73 billion by 2031 at a compound annual growth rate (CAGR) of 18.9% during the forecast period, according to a recent report from Growth Plus Therapeutics. Note that there were no real treatments, only one protective antibody, Synagis, available in 2022.&nbsp;
The therapeutic used for treating RSV is Ribavirin, a highly toxic compound also used as a therapy for hepatitis C. However, Ribavirin carries with it some severe side effects, including the destruction of red blood cells, leading to liver, kidney and spleen toxicities and failures &ndash; which limit its use to patients with a high risk of progressively severe RSV. The primary concerning side effect of Ribavirin is anemia, a decrease in red blood cells that can cause a drop in hemoglobin. Ribavirin-induced anemia can manifest within two weeks of starting the therapy and can get worse as the drug dosage increases.
<h2>NanoViricides Treatment For RSV</h2>
NanoViricides (NYSE American: <a class="ticker" href="https://www.benzinga.com/stock/nnvc#NYSE American">NNVC</a>), a leading developer of antiviral treatments through its nanomedicines platform, is developing a safe and effective alternative therapeutic for RSV. Its drug candidate, NV-387, has already been shown to not have the toxicity or side effects <a href="https://www.benzinga.com/pressreleases/23/07/r33186327/companys-broad-spectrum-antiviral-nv-387-has-demonstrated-excellent-effectiveness-in-rsv-in-a-leth">associated with Ribavirin</a>, according to the company. It reports that in the recent pre-clinical animal study, NV-387 almost matched the efficacy of Ribavirin with no toxicity. This animal study was designed to mimic the lethal lung pneumonia in infants caused by RSV infection.&nbsp;
Importantly, NV-387 is already in phase 1 human clinical trials as the active ingredient in the company&rsquo;s COVID drug candidate NV-CoV-2. The company has just reported that it was found to be completely safe, with no side effects found to date, in this continuing phase 1 Study. For this reason, the company anticipates soon being able to advance NV-387 to a phase 2 human clinical trial against RSV.&nbsp;
If the results of animal studies are replicated in humans, this nanoviricides drug could become truly a game-changer in the treatment of RSV infections.&nbsp;

Also, Remdesivir can only be administered as an infusion in a hospital, limiting the patient populations who can get access to the drug.&nbsp;
Two other drugs, Molnupiravir by Merck &amp; Co. (NYSE: <a class="ticker" href="https://www.benzinga.com/stock/mrk#NYSE">MRK</a>) and Paxlovid by Pfizer (NYSE: <a class="ticker" href="https://www.benzinga.com/stock/pfe#NYSE">PFE</a>) are both orally available. The WHO has issued a strong recommendation for Paxlovid, but only a conditional recommendation for Molnupivavir for patients with severe COVID-19 or non-severe COVID-19 at the highest risk of hospitalization.

(NYSE: <a class="ticker" href="https://www.benzinga.com/stock/mrk#NYSE">MRK</a>), Paxlovid from Pfizer Inc. (NYSE: <a class="ticker" href="https://www.benzinga.com/stock/pfe#NYSE">PFE</a>) and Remdesivir from Gilead Sciences, Inc. (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/gild#NASDAQ">GILD</a>). However, there are limitations or drawbacks to each of these treatments.&nbsp;
As a broad-spectrum anti-coronavirus drug, NV-CoV-2 has been shown to work against <a href="https://www.benzinga.com/general/biotech/23/03/31608814/covid-remains-a-global-threat-nanoviricides-set-to-begin-clinical-trials-for-pan-coronavirus-drug">a variety of unrelated coronaviruses</a>. This suggests that different strains or mutations are unlikely to escape the drug. Several common coronavirus strains include hCoV-229E, hCoV-OC43, hCoV-HKU1, and hCoV-NL63, which recur seasonally. Of these, NL63 and OC43 present with pneumonia similar to the delta COVID variant in susceptible patients, leading to fatalities at rates that are comparable to seasonal influenza. Now, SARS-CoV-2 with its variants is added to this list as a commonly occurring coronavirus.&nbsp;
<h2>A Nanomedicine Platform For Other Viruses</h2>
The trials for NV-CoV-2 are part of the company&rsquo;s development of a novel platform technology employing nanoviricides. The NV-CoV-2 drug contains NV-387, a first-in-class chemical nanomedicine, and its unique mechanism of action (MoA) doesn&rsquo;t rely on the patient&rsquo;s immune system to destroy the virus. The Company says that NV-387 mimics an attachment receptor family to which <a href="https://profiles.smallcapsdaily.com/nnvc/">over 90% of known viruses</a> bind, and NanoViricides hopes that NV-387 will likely be a clinically effective drug against at least a few of these viruses. The Company has said that it has begun R&amp;D work to expand the indications of NV-387 to other viruses.&nbsp;
By bringing NV-CoV-2 to human trial, NanoViricides has demonstrated the platform&rsquo;s capability to deliver a clinically relevant antiviral. After the Phase 1 studies are completed, the safety and tolerability data will be applicable to any other viruses that the Company targets using NV-CoV-2, assuming it can demonstrate pre-clinical success. Then NV-387 can directly enter Phase 2 as an antiviral against such additional viruses &mdash; which would likely mean significant cost and time savings for the additional indications, and substantial improvement in the return on investment (ROI) for the Company with commensurate benefits to the investors.&nbsp;
The Company is not a &ldquo;one-trick pony&rdquo; either. It already has NV-HHV-1, a drug to treat <a href="https://www.nature.com/articles/d43747-020-00242-w">shingles</a> rashes, that has completed pre-clinical studies. The company believes its platform could provide treatments for a host of viruses, including other herpesviruses (cold sores, genital ulcers, and many other serious diseases), as well as influenza and HIV.
This would position NanoViricides at the intersection of multiple growing treatment markets. The herpes treatment market was worth <a href="https://www.grandviewresearch.com/industry-analysis/herpes-simplex-virus-treatment-market-report#:~:text=Report%20Overview,9.24%25%20from%202023%20to%202030.">$1.5 billion in 2022</a> and is predicted to grow at a compound annual growth rate (CAGR) of 9.2% over the next seven years. The influenza market is expected to reach <a href="https://www.alliedmarketresearch.com/influenza-treatment-market-A06106">$1.2 billion by 2026</a>, while the HIV market is predicted to reach <a href="https://www.alliedmarketresearch.com/hiv-drugs-market">$36 billion by 2027</a>.&nbsp;&nbsp;
For now, however, the company is keeping laser-focus on the development of NV-CoV-2 for treating COVID-19 and long COVID. Its current trial will test the drug&rsquo;s safety and efficacy, and the data will be used to prepare for phase 2 trials.

Novel Nanomedicine Platform With Pipeline Of Antivirals Goes To Human Trials – What Other Viruses Could This COVID Treatment Target?

NanoViricides, Inc. (NYSE American:&nbsp;<a href="https://www.bloomberg.com/quote/NNVC:US">NNVC</a>) (the &ldquo;Company&rdquo;) reports that the clinical trial of its broad-spectrum antiviral drug NV-CoV-2 is progressing satisfactorily.&nbsp;
NanoViricides is a clinical-stage global leader in the development of highly effective antiviral therapies based on a novel nanomedicines platform. NV-CoV-2 (API NV-387), our lead drug candidate for the treatment of coronavirus infections including COVID and potentially many cases of long COVID, is in Phase1a/1b Safety and Preliminary Efficacy Human Clinical Trials initiated by the Drug Sponsor Karveer Meditech Pvt. Ltd. India, the Company&rsquo;s Licensee and co-developer in India.
Following safety and tolerability evaluation in healthy persons for a single escalating dose of NV-CoV-2 Oral Syrup or NV-CoV-2 Oral Gummies in the Part 1a, the clinical trial will continue into Part 1b if there are no serious adverse events.&nbsp;
Phase 1a is Progressing Rapidly: 
Enrollment in the third and highest single dose level of 40mg/Kg NV-CoV-2 Oral Syrup, and separately, 2,000mg NV-CoV-2 Oral Gummy has already begun. The lowest dose cohorts in the clinical trial (10mg/Kg Oral Syrup, and separately, 500mg Gummy) have completed, and the middle dose cohorts (20mg/Kg Oral Syrup, and separately, 1000mg Gummy) have been substantially completed allowing the highest dose cohorts to begin. Each person after dosing is under observation (in-hospital stay) for 48 hrs, followed by a scheduled follow-up visit.&nbsp;
There were no adverse events to date at any of the dose levels including the highest dosages.&nbsp;
Phase 1b to Begin Shortly: 
In Phase 1b, healthy persons will be dosed with multiple doses of the Oral Syrup and separately, Oral Gummies to study Safety and Tolerability.&nbsp;
Additionally, in Phase 1b, in separate cohorts, patients with mild to moderate/severe COVID-19 shall be enrolled to assess indication of efficacy. Patients deemed by the physician to be likely to require hospitalization within 48 hrs of screening will be excluded.&nbsp;
&ldquo;We are pleased with the success of the clinical trial so far and look forward to the start of the Phase 1b portion soon,&rdquo; said Anil R. NV-387 is designed as a bio-mimetic that can possibly be an effective drug against many viruses including the coronaviruses. &ldquo;Resistance is Futile&rdquo;: NV-387, the active pharmaceutical ingredient of NV-CoV-2 is designed to mimic a cell membrane with a number of so called &ldquo;attachment receptor sites&rdquo; chemically covalently connected to each polymer chain in the nanomicelle. No matter how much a virus changes, it still binds to the same attachment receptor(s), and therefore, it is unlikely to escape the nanoviricide drug.&nbsp;
This design we believe solves the major issue of small molecule as well as antibody therapeutics, namely, development of resistant virus variants.&nbsp;
NV-CoV-2 is Aimed at Satisfying Many Unmet medical Needs in COVID: NV-CoV-2 was shown to be extremely safe in pre-clinical animal studies. It was also found to be extremely effective in lethal infection animal model studies.&nbsp;
Thus we believe that NV-CoV-2 will be useable in all segments of patient populations, (i) in age from pediatric to geriatric, with otherwise healthy adults included; (ii) with or without co-morbidities; (iii) with disease manifestation from mild, moderate, severe to hospitalized stage.&nbsp;
In contrast, existing COVID therapeutics are limited in the treatable segment(s) of population; thus, Remdesivir is indicated for hospitalized patients only; Molnupiravir and Paxlovid are both indicated for patients over 65 years of age with co-morbidities that are not taking other drugs that would cause interactions. This leaves a large patient population that is unserved.&nbsp;
Further, we believe that NV-387 may become an important drug for the treatment of certain cases of long COVID wherein residual virus is known to be present.&nbsp;
NV-387 May Have a Very Large Range of Indications, because Over&nbsp;90%&nbsp;of All Human Viruses Use the Attachment Receptor(s) Mimicked by NV-387: 
NV-387, the active pharmaceutical ingredient of NV-CoV-2, mimics a family of attachment receptors called sulfated proteoglycans (S-PG), or glycosaminoglycans (GAGs). This family includes heparan sulfate (HSPG), dermatan sulfate&nbsp; (DSPG), chondroitin sulfate (CSPG), and keratan sulfate (CSPG). Over&nbsp;90%&nbsp;of known pathogenic viruses bind to one or more of these attachment receptors. These viruses include Coronaviruses, Paramyxoviruses (RSV - Respiratory Syncytial Virus, and HMPV- human MetaPneumoVirus), Dengue Viruses, HerpesViruses, Human PapillomaViruses (HPV), HIV, Hendra and Nipah Viruses, Ebola and Marburg Viruses, among others.&nbsp;
NV-387 is likely to be effective as a clinically viable drug candidate against at least some of these viruses, we believe. Many of these viruses have no available antivirals or have antivirals with limited applicability.&nbsp;
We have already undertaken a program to expand the potential indications of NV-387. Success in any of these studies would enable direct entry into Phase II/III clinical trials for that indication.&nbsp;
Such expansion of use of NV-387 would significantly expand the market size and substantially improve the return on investments (ROI).&nbsp;&nbsp;

In 2024, NanoViricides aims to <a href="https://ca.finance.yahoo.com/quote/NNVC/">continue advancing NV-CoV-2</a> through further clinical trials, expanding its treatment capabilities to address unmet medical needs.
