Intellia Therapeutics, Inc. (NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/NTLA#NASDAQ">NTLA</a>), a leading clinical-stage gene editing company focused on revolutionizing medicine with CRISPR-based therapies, today announced that the U.S. Food and Drug Administration (FDA) has granted Regenerative Medicine Advanced Therapy (RMAT) designation to nexiguran ziclumeran (nex-z, also known as NTLA-2001) for the treatment of hereditary transthyretin (ATTR) amyloidosis with polyneuropathy (ATTRv-PN). Nex-z is an in vivo&nbsp;CRISPR-based investigational&nbsp;therapy designed as a single-dose treatment to inactivate the&nbsp;TTR&nbsp;gene and thereby prevent the production of TTR protein for the treatment of ATTR amyloidosis. Development and commercialization of nex-z is led by Intellia as part of a multi-target collaboration with Regeneron.
"This RMAT designation underscores the transformative potential of nex-z, our investigational in vivo CRISPR-based gene editing therapy for those living with hereditary ATTR amyloidosis with polyneuropathy," said Intellia President and Chief Executive Officer John Leonard, M.D. "It was granted following the FDA's review of our compelling interim Phase 1 data that indicated our one-time treatment led to rapid, deep and durable TTR reduction, which is expected to halt and potentially reverse the disease. The RMAT designation was established under the 21st Century Cures Act to expedite the development and review of promising therapeutic candidates, including genetic therapies, that are intended to treat, modify, reverse or cure a serious or life-threatening disease. RMAT designation includes benefits, such as early interactions with the FDA, including discussions on surrogate or intermediate endpoints that could potentially support accelerated approval and satisfy post-approval requirements, and potential priority review of a product's biologics license application (BLA).
This RMAT designation is the third special regulatory designation received by Intellia for nex-z. Nex-z was also granted Orphan Drug Designation by the U.S. FDA and European Union Orphan Drug Designation by the European Commission.
About nexiguran ziclumeran (nex-z, also known as NTLA-2001) Based on Nobel Prize-winning CRISPR/Cas9 technology, nex-z has the potential to become the first one-time treatment for transthyretin (ATTR) amyloidosis. Nex-z is designed to inactivate the&nbsp;TTR&nbsp;gene that encodes for the transthyretin (TTR) protein. Interim Phase 1 clinical data showed the administration of nex-z led to consistent, deep and long-lasting TTR reduction. The co-primary endpoints were the change in cognition and function from baseline to the end of the double-blind treatment period at week 52, assessed by the ADAS-COG12 and ADCS-ADL scales, comparing simufilam to placebo. Simufilam continued to demonstrate an overall favorable safety profile. The Company will hold a webcast today at 8 AM ET.
"The results are disappointing for patients and their families who are living with this disease and physicians who have been looking for novel treatment options. We took careful measures to enroll patients with mild-to-moderate AD. Despite that, the loss of cognition in the placebo group was less pronounced than was previously reported in other placebo-controlled studies in AD. We are working to understand this better," said Rick Barry, President and Chief Executive Officer. "A result like this has implications on our second Phase 3 trial,&nbsp;ReFocus-ALZ. We have made the difficult decision to discontinue ReFocus-ALZ, given the nature of today's reported results. The complete 52-week dataset will be available from the study along with a large portion of 76-week data. We intend to report detailed analyses of both studies in the future. We will also be discontinuing the Open Label Extension study."
Mr. Barry continued, "We have a special gratitude for the patients and their families and caregivers who participated in our clinical program for AD. We are also immensely grateful to our employees, study investigators and site coordinators, as well as our other partners, for their commitment to this program. MicroStrategy Inc. (NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/MSTR#NASDAQ">MSTR</a>) has been described as the &#8220;Bitcoin magnet,&#8221; attracting billions of dollars in global capital <a href="https://www.benzinga.com/markets/cryptocurrency/24/11/42138209/microstrategy-trading-at-insane-256-premium-to-bitcoin-holdings-research-shows">through its unprecedented Bitcoin</a> (CRYPTO: <a class="ticker" href="https://www.benzinga.com/quote/btc/usd">BTC</a>) treasury strategy. 
What Happened: Bernstein's report on Monday highlights MicroStrategy's role in mainstreaming Bitcoin on Wall Street, projecting the company could hold 4% of the global Bitcoin supply by 2033, up from 1.7% today.
However, skeptics, including popular analyst &#8216;Kryptokrates,&#8217; caution that the strategy comes with significant risks, raising questions about its long-term viability.
Bernstein lauded MicroStrategy's innovative model, which <a href="https://www.benzinga.com/24/11/42151809/michael-saylor-reveals-microstrategy-is-making-500m-a-day-with-bitcoin-may-very-well-be-the-most-profitable-company-in-the-us-growing-the-fastest-right-now">leverages convertible debt</a> and equity raises to amass Bitcoin.
The firm's current holdings of ~300,000 BTC are valued at $32 billion, acquired at an average cost of ~$49,900 per Bitcoin. 
Bernstein projects MicroStrategy could increase its holdings to 4% of the world's Bitcoin by 2033, assuming Bitcoin reaches $1 million per coin.
"MicroStrategy's Bitcoin treasury model is unprecedented," Bernstein noted. "A company on an insatiable path to attract billions in global capital to invest in Bitcoin."
MicroStrategy has raised more than $7 billion in convertible debt, with a long-term plan to secure $42 billion in capital by 2027. 
<a href="https://www.benzinga.com/markets/cryptocurrency/24/11/42138209/microstrategy-trading-at-insane-256-premium-to-bitcoin-holdings-research-shows">Its approach</a> involves adding Bitcoin to the balance sheet, increasing stock volatility and using convertible debt to enable low-cost capital raises.
Bernstein sees this strategy as <a href="https://www.benzinga.com/24/11/42151809/michael-saylor-reveals-microstrategy-is-making-500m-a-day-with-bitcoin-may-very-well-be-the-most-profitable-company-in-the-us-growing-the-fastest-right-now">sustainable</a> under favorable market conditions, raising its price target for the stock to $600, a 42% upside from its current $421.88.

Institutional Appeal vs. Retail Risks
While Bernstein remains bullish, critics like Kryptokrates argue that MicroStrategy's financial engineering introduces systemic risks.
"Saylor created the perfect financial perpetual motion machine. It works until it doesn't. The question isn't if, but when,&#8221; the analyst <a href="https://x.com/Kryptokrates/status/1860381306103746766">tweeted</a>.
Kryptokrates outlined several potential vulnerabilities:

<li>Leverage Concerns: "MicroStrategy's total debt is ~$3 billion—a massive bet on ‘number go up.'"</li>
<li>Regulatory Pressure: "Any regulatory crackdown could disrupt the model."</li>
<li>Retail Investor Risk: "Small investors think they're ‘buying Bitcoin exposure' but are actually buying systemic risk."</li>
</ol>
Kryptokrates also flagged warning signs such as increasing convertible note issuances and more aggressive Bitcoin buying, suggesting heightened financial risk as the company scales.
Bernstein emphasized the appeal of <a href="https://www.benzinga.com/markets/cryptocurrency/24/11/42121936/bitcoin-boom-20-microstrategy-gains-traction-as-a-leveraged-bet-on-crypto-growth">MicroStrategy's strategy</a> for institutional investors, citing high liquidity, Bitcoin exposure, and regulatory compliance.
Kryptokrates, however, noted that retail investors might misunderstand the risks, tweeting, "Nothing is free in finance. Leverage works both ways. Always check the fine print."
The best-case scenario, according to Bernstein, sees MicroStrategy continuing to capitalize on Bitcoin's price appreciation, reaching $1 million per coin by 2033. 
However, Kryptokrates offered a stark contrast: "Worst case—Bitcoin crashes, notes demand repayment, share price collapses, system fails."
