Edible coatings are commonly used to enhance the quality and increase the shelf life of fruits and vegetables. However, industrial-scale use of edible coatings has struggled with effectiveness limitations due to poor stability and solubility of the coatings. Recent developments in nanoemulsion-based coating technology offer promise for an effective and efficient strategy but have been challenged by industrial-scaling limitations as well as quality and stability inconsistencies from current nanoemulsion technology platforms.

For the brands that originally benefited from that exclusivity, the so-called patent cliff represents a painful transition period as their revenue takes a hit and they&rsquo;re forced to find other ways to make up the losses. But for small and mid-sized biotechs and pharmaceutical companies, in particular, the patent cliff is a chance to develop a biosimilar competitor to these previously off-limits drugs.&nbsp;

In 2018, Jazz Pharma&rsquo;s (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/jazz#NASDAQ">JAZZ</a>) Epidiolex, a seizure medication, became the first and only cannabis-derived prescription drug to receive FDA approval. Since then, more and more clinical trials are underway to develop other therapeutics tapping the plant&rsquo;s medicinal uses, but approval has been painfully slow because complex regulations added so many extra steps and redundancies.&nbsp;
That weedy regulatory landscape changed last year when President Biden signed the <a href="https://www.arnoldporter.com/en/perspectives/advisories/2022/12/medical-cannabis-research">Medical Marijuana and Cannabidiol Research Expansion Act </a>into law in December. Drug developers will no longer need to seek separate approval from the Drug Enforcement Agency (DEA). It also enforces shorter DEA registration and review processes and tasks the agency with ensuring that there is an adequate and uninterrupted supply of marijuana for researchers working on new therapeutics.&nbsp;
Earlier that same year, Jazz began construction on a <a href="https://www.fiercepharma.com/pharma/jazz-and-gw-pharma-grow-cannabinoid-manufacturing-ambitions-new-100m-plant">$100 million manufacturing facility</a> that will be dedicated to producing cannabis-based medicines. PBI&rsquo;s UltraShear&trade; platform is one of the highest pressure processing homogenizing technologies in the world today. That ultra-high pressure accomplishes what other manufacturers have been trying to do with chemicals or heat. It slices through oil, breaking it down into nano-sized oil droplets that are so small, they are effectively water-soluble. The 30-milliliter bottle contains 40 milligrams of CBD oil per milliliter. But what makes the product such a game changer in the industry is that it&rsquo;s manufactured using PBIO&rsquo;s patented UltraShear&trade; technology. The pressure-based platform is uniquely designed to break down oil molecules using pressure rather than heat or chemicals that could alter or damage the product. Above all, the conversion is a key signal of growing investor confidence as the deal was consummated at a value of $2.50 per share, which is about 38% higher than the stock&rsquo;s current open market price. PBI President and CEO Richard T. <h2>PBI Continues Expansion Via Collaboration Agreements Across Multiple Industries</h2>
That bullish sentiment is largely driven by a series of recent milestones PBI has achieved over the past few months as the company works to transition into profitability and uplist to NASDAQ/NYSE. Earlier this month, Pressure BioSciences, Inc. (OTCQB: <a class="ticker" href="https://www.benzinga.com/stock/pbio#OTCQB">PBIO</a>) <a href="https://www.benzinga.com/pressreleases/23/01/ac30480016/pressure-biosciences-nanoemulsions-platform-forging-game-changing-future-for-cannabis-industry">announced</a> potentially ground-breaking results from a consumer focus group testing a THC oral spray developed using the company&rsquo;s innovative UltraShear&trade; Technology nanoemulsification platform. The oral spray boasted strong potency and extremely fast absorption that could put it on track to become a best-in-class cannabis product in terms of speed to effects, stability, and bioavailability.&nbsp; The THC oral spray tested in this consumer focus group is being developed in partnership with California-based cannabis manufacturer, Cr&egrave;me De Canna.
The news comes on the heels of the company&rsquo;s December <a href="https://www.benzinga.com/pressreleases/22/12/ac30060435/pressure-biosciences-announces-first-commercial-production-of-revolutionary-ultrashear-nanoemulsi">announcement</a> of its first commercial shipment of a CBD oral spray manufactured with the same UltraShear technology. The $75,000 shipment to Safer Medical of Montana is the first of many shipments the company expects to make to distributors of CBD products throughout the U.S. during 2023. In recent press releases, PBIO has stated that CBD products manufactured using its patented UltraShear platform could potentially yield many millions in revenue before the end of its 2023 fiscal year.&nbsp;
<h2>Slow And Inconsistent THC Absorption Remains A Key Challenge for Cannabis Manufacturers</h2>
Because cannabinoids, like the well-known THC and CBD, are oil-soluble rather than water-soluble, the human body struggles to absorb them efficiently. As a consequence, much of the active ingredient in edible cannabis products is absorbed slowly, intermittently, or hardly at all.&nbsp;
In one <a href="https://pubmed.ncbi.nlm.nih.gov/16237477/">review</a> of cannabis research, only 4% to 12% of THC was absorbed by the body when taken orally. Not only does that mean that the majority of the THC in an edible ends up getting wasted, but the variation in bioavailability between 4% and 12% is highly significant. A consumer eating the same edible product from the same manufacturer with the same dose might absorb just 4% one day but 12% the next day, leading to very different results.&nbsp;
This makes it incredibly difficult for manufacturers to achieve precise dosing and effects in their edibles. It also increases production costs since they often need to include more than the minimum effective dose to compensate for the fact that most of the active ingredients won&rsquo;t be absorbed.
In addition to inefficient and inconsistent cannabinoid absorption, another challenge in the edible cannabis industry is absorption time. It is estimated to take anywhere from <a href="https://www.ccsa.ca/sites/default/files/2019-06/CCSA-Cannabis-Inhaling-Ingesting-Risks-Infographic-2019-en_1.pdf">30 minutes to two hours</a> from the time a consumer ingests an edible to the time they start to feel its first effects, and then up to four hours to achieve peak effects.&nbsp;
&ldquo;Impatient consumers may repeatedly consume their edibles, only to be unhappily overdosed shortly thereafter,&rdquo; explained PBI&rsquo;s Director of Sales and Marketing, John Hollister in a <a href="https://www.benzinga.com/pressreleases/23/01/ac30480016/pressure-biosciences-nanoemulsions-platform-forging-game-changing-future-for-cannabis-industry">statement</a> on the consumer testing results. For patients using cannabis as a treatment for pain, anxiety, or depression, that delayed response also means edible cannabis is often unable to provide the instant relief they need.&nbsp;
While smoking or vaping comes with a higher bioavailability of around 30% and a faster response time of seconds or minutes, the respiratory health consequences make it a less-than-ideal option, especially for regular use.&nbsp;
<h2>PBI&rsquo;s UltraShear Nanoemulsion Technology Could Be A Breakthrough In Achieving Precision Dosing for THC, CBD, and Other Products</h2>
Consumers in the focus group reported that the first effects could be felt less than 10 minutes after initially using the innovative UltraShear processed THC oral spray, with many reporting noticeable effects within 1-5 minutes. Peak effects were achieved between 15-60 minutes after taking the dose. The results of this initial focus group could be a game-changer for an industry that&rsquo;s been struggling with the challenges around consistent dosing and effective non-inhalation absorption.&nbsp;
&ldquo;This testing has provided dramatic consumer-level validation in the cannabis market for our UltraShear nanoemulsion capabilities and for its expected market-changing impact,&rdquo; said Mr. Hollister.&nbsp;
The UltraShear technology used to make the oral spray is PBI&rsquo;s patented pressure-based platform that&rsquo;s capable of creating what the company describes as the &ldquo;best-in-class&rdquo; nanoemulsions worldwide. In the cannabis nanoemulsion, the oil drops containing the CBD or THC molecules are sheared down to a nanosize so small that they are effectively water-soluble. This makes for a significantly more easily absorbed product that can be more precisely and accurately dosed.
For consumers, that means more immediate and consistent effects every time they use an UltraShear processed oral spray or other mechanisms for ingestion. For manufacturers, that improved bioavailability means smaller amounts of THC or CBD are required to achieve an effective dose since the less active ingredient is getting wasted - thus saving the manufacturer money.

The pressure-based equipment is reportedly uniquely capable of producing nanoemulsions &mdash; an emulsion with oil droplets so small (nano) they can&rsquo;t recombine and separate from the rest of the mixture like standard emulsions do. With demonstrations expected to begin at Ohio State <a href="https://ir.pressurebiosciences.com/press-releases/detail/390/pressure-biosciences-updates-stakeholders-with-solid">this summer</a>, UST processing&nbsp; could have major implications for food and beverage companies and businesses in the health and wellness space.
<h2>PBI And Ohio State Partner To Develop And Explore Commercial Uses Of UST Platform</h2>
PBI&rsquo;s UST platform for nano emulsification of oil-based products, like CBD oil or Vitamin D3 oil, allows a manufacturer to &ldquo;destroy bacteria and other pathogens, which in turn may reduce or eliminate the need for chemical (anti-microbial) additives, thus increasing food safety and quality,&rdquo;&nbsp; said PBI Vice President Dr. Edmund Ting, who is the head of engineering at PBIO, and co-investigator of the U.S. Department of Agriculture fundedUST project with Ohio State.
Beyond removing bacteria and other pathogens, UST-processed nanoemulsions can also be more stable, meaning manufacturers won&rsquo;t have to rely on additives to stop the mixture from separating as they do in larger emulsions like salad dressings or natural peanut butter.&nbsp;
&ldquo;It&rsquo;s not just drinks that might soon be preserved in a much healthier way, as UST can also be used by food manufacturers in the healthier processing of sauces, condiments and other liquid foods, including nutritional drinks, ice cream mix, juices and food emulsions,&rdquo; said Professor V.M. &ldquo;Bala&rdquo; Balasubramaniam, an expert in food processing who heads the project at Ohio State.
PBI also reported that because the nano-sized oil droplets are not only more stable but also more readily absorbed by the human body, UST processing could also improve the bioavailability of oil-based nutrients like antioxidants (Astaxanthin) and cannabidiol (CBD) that are typically difficult for the body to absorb because they are contained in oil.
In studies using this novel&nbsp; platform, UST successfully <a href="https://ir.pressurebiosciences.com/press-releases/detail/368/pressure-biosciences-novel-ust-platform-delivers/privacy-policy">processed Astaxanthin</a>, one of the world&rsquo;s most potent antioxidants, into a more bioavailable, shelf-stable nanoemulsion that can then be easily infused into beverages or or liquid foods. In December, its CBD oil nanoemulsion demonstrated a record-breaking <a href="https://ir.pressurebiosciences.com/press-releases/detail/384/pressure-biosciences-shatters-preconceived-nanoemulsion">18-months of shelf stability</a> at room temperature.
<h2>PBI Expects Commercial Scale UST Platform To Reach The Market This Year&nbsp;&nbsp;</h2>
The <a href="https://ir.pressurebiosciences.com/press-releases/detail/313/pressure-biosciences-announces-achievement-of-first-major">installation at OSU</a> earlier this month represents the first commercial-scale version of PBI&rsquo;s technology. The high-pressure processing (HPP) food market is a $14 billion market that encompasses the technology used to sanitize, pasteurize or otherwise improve the safety and quality of food and beverages.&nbsp;
<h2>How Does Traditional High-Pressure Processing Work?</h2>
HPP involves using ultra high pressure to reduce the level of microorganisms in liquid foods and beverages; such pathogens could be harmful - even deadly - to people if ingested. By applying uniform pressure to the entire item, HPP pasteurization systems offered by industry players like JBT Avure Technologies (NYSE: <a class="ticker" href="https://www.benzinga.com/stock/jbt#NYSE">JBT</a>), Thyssenkrupp ADR (OTC: <a class="ticker" href="https://www.benzinga.com/stock/tkamy#OTC">TKAMY</a>), and Hyperbaric are able to destroy food-borne pathogens and inactivate deleterious enzymes without harming the food itself.&nbsp;&nbsp;
Before HPP came on the scene, sanitizing food required either high heat or strong chemical preservatives. Both of these worked in terms of killing off harmful pathogens, but they would also adversely impact the flavor, texture, color, and nutritive value of the food. Even less desirable, they could also end up destroying certain nutrients &mdash; especially Vitamin C and other antioxidants that break down in heat, reducing the nutritional value of the food.
HPP, on the other hand, doesn&rsquo;t change the texture or flavor of the food itself as much as heat or chemicals do. Plus, using pressure instead of heat also helps preserve the nutrient profile since it doesn&rsquo;t break down vitamins, antioxidants, or other micronutrients in the way that high heat does.&nbsp;
UST also allows manufacturers to achieve longer shelf life without relying on chemical preservatives &mdash; and in a market where consumers are increasingly seeking &ldquo;clean label&rdquo; foods and are generally more wary of any kind of additives, HPP makes it possible to appeal to evolving consumer demands.
Despite its benefits, existing HPP methods can&rsquo;t overcome a key problem that liquid food and beverage manufacturers face: preventing emulsions from separating. With many foods, especially those that combine oil with water, the liquids don&rsquo;t want to stay mixed as the product sits on the shelf, so the oil tends to separate and rise to the top.
Some manufacturers deal with this by using hydrogenated fats to create foods that won&rsquo;t separate, but these processed fats are associated with a possible <a href="https://www.mayoclinic.org/diseases-conditions/high-blood-cholesterol/in-depth/trans-fat/art-20046114">increased risk</a> of heart attacks, diabetes, and stroke. When manufacturers skip the hydrogenated fats and just let separation happen, the desired health benefits of the food is often significantly reduced, since the human body struggles to absorb oil-based nutrients.&nbsp;
<h2>How Does Pressure BioSciences&rsquo; UST Platform Overcome the HPP Shortcomings?</h2>
The UST Platform is a pressure-based system that might finally solve the problem of separation in emulsions, without using chemical processing like hydrogenation.
Where existing HPP technology relies on pressure alone, the UST platform combines ultra-high pressure levels with a patented NanoGap valve that controls flow rates in a novel way for better dispersion.&nbsp; Adding this NanoGap valve-related shear technology (or the ability to physically break apart and disperse the oil drops) is a game-changer.&nbsp;
The unique design of the NanoGap valve makes it possible to produce nanoemulsions where oil drops are broken down into tiny oil droplets that are so small they can&rsquo;t easily recombine, which makes the resulting nanoemulsion more shelf-stable, meaning manufacturers can ship products worldwide without worrying about separation or quality degradation. In addition to being significantly more stable, the resulting nanoemulsions offer the additional advantage that the active oil-based ingredient is likely to be more bioavailable, meaning the body absorbs more of the oil-based active ingredient.&nbsp;&nbsp;
Not only could the UST process make it possible to eliminate hydrogenated fats from store shelves, but it may also make previously difficult-to-absorb supplements like CBD oil or the powerful antioxidant Astaxanthin easier to infuse into beverages and liquid foods.&nbsp; Importantly, the UST platform can also scale to very large manufacturing levels.
Essentially, UST-derived nanoemulsions not only protect against nutrient loss by significantly reducing exposure to heat, but they can also improve the bioavailability of those oil-based nutrients while reducing manufacturers&rsquo; dependence on chemical additives and chemical processing that often increase the cost of production while resulting in adverse health effects.
To make the UST platform more widely available in the food and beverage space, PBI partnered with The Ohio State University (OSU) to develop a commercial-scale model of the UST technology. Now on display at the world-renowned Advanced Food Technology Pilot Plant at Ohio State, the model system will soon be available to provide live demonstrations of the nanoemulsification process for industry companies from around the world.&nbsp;
