Shares of Alterity Therapeutics (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/athe#NASDAQ">ATHE</a>) were up 7.8% on Jul 17 after the company announced positive interim data from the phase II ATH434-202 study evaluating its lead pipeline candidate ATH434 for treating patients with multiple system atrophy, a rare neurodegenerative disease. The study enrolled 10 participants with advanced MSA.
Per the press company, interim analysis results from the open-label ATH434-202 study included clinical and biomarker data from seven participants who were treated with ATH434 for six months and neuroimaging data from three participants who were treated with ATH434 for 12 months.
Data from the same showed that 43% of the participants displayed improvement on the Unified MSA Rating Scale Part I (UMSARS1), indicating reduced disability in activities of daily living following six months of treatment. Also, 29% of the participants had stable or improved neurological symptoms at the same time.
Patients who were treated with ATH434 had favorable clinical and biomarker outcomes, which underlined the potential of ATH434 to modify the course of MSA treatment.
Shares of Alterity have plunged 14.2% so far this year compared with the industry&#39;s decline of 1%.
Phase I studies conducted on ATH434 showed that the candidate is well tolerated and achieved brain levels that are comparable to efficacious levels seen in animal models of MSA.

Shares of Lexeo Therapeutics (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/lxeo#NASDAQ">LXEO</a>) lost 26% on Monday after it reported positive interim data from two cohorts of the phase I/II SUNRISE-FA study evaluating LX2006, an investigational gene therapy for treating Friedreich ataxia (FA) cardiomyopathy.
Data from the SUNRISE-FA study showed that treatment with LX2006 achieved a mean reduction in left ventricular mass index (LVMI) of 11.4% at 12 months and 18.3% at 18 months in participants with elevated LVMI at baseline. LVMI is an estimator of cardiac morbidity and mortality. Treatment with the therapy was also well-tolerated across all study participants, with no treatment-related serious adverse events.
The interim data also showed that treatment with LX2006 also showed improvement in other key measures of cardiac status, including left ventricular wall thickness (an early indicator of an inefficient heart) and troponin I (a biomarker of myocardial injury), in the majority of participants at 12 months.
Based on the above results, management sees a path for seeking the FDA&#39;s approval for LX2006 under the accelerated pathway.
Lexeo intends to report additional data at a future medical meeting later this year. Management is currently dosing patients in the third dose cohort of the SUNRISE-FA study, which is also enrolling study participants.
While Wall Street analysts were impressed with the results, some investors raised concerns about the data. They stated that the results were not enough to warrant accelerated approval from the FDA. The data also did not show that patients&#39; function improved after treatment with LX2006. This was likely the reason for the share price decline.
Year to date, Lexeo&#39;s shares have lost 3.0% compared with the industry&#39;s 3.6% fall.

uniQure N.V.&nbsp;(NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/qure#NASDAQ">QURE</a>) surged 76.5% on Jul 9 after it announced updated interim data, including up to 24 months of follow-up data, from 29 treated patients&nbsp;enrolled in the ongoing early to mid-stage studies of AMT-130 for Huntington&#39;s disease in the United States and EU. Results demonstrated a potential long-term, durable clinical benefit and reduction of a key marker of neurodegeneration upon treatment with the candidate.
AMT-130, a one-time administered investigational gene therapy, is being developed to treat Huntington&#39;s disease. Huntington&#39;s disease is a genetic disorder that causes the progressive breakdown of nerve cells in the brain, which leads to a decline in cognitive and physical abilities, often resulting in movement, thinking and psychiatric problems.
uniQure is simultaneously conducting two phase I/II studies of AMT-130, with 26 participants in the United States and 13 in the EU/UK to treat Huntington&#39;s disease. Among these, a total of 29 patients received either a low dose (12 patients) or a high dose (17 patients) of AMT-130, while 10 control patients underwent imitation surgery. As of Mar 31, 2024, follow-up data at 24 months were available for 21 patients, including 12 from the low-dose group and nine from the high-dose group.
Please note that uniQure, for the first time, performed a statistical analysis of clinical outcomes at 24 months for the 21 treated patients. This analysis, compared with an expanded, propensity-weighted external control group of 154 patients, developed in collaboration with the Cure Huntington&#39;s Disease Initiative. The control data was derived from the TRACK-HD, TRACK-ON and PREDICT-HD natural history studies.
Year to date, shares of QURE have lost 1.5% compared with the industry&#39;s 6.3% decline.

Per the data readout from the phase I/II Huntington&#39;s disease studies, patients treated with the high dose of AMT-130 experienced a statistically significant, dose-dependent slowing in disease progression of 80%, as measured by cUHDRS after 24 months. For the low-dose group, a 30% slowing of disease progression, as measured by cUHDRS, was observed compared with the control group at month 24.
Please note that the cUHDRS has been demonstrated to be the most sensitive measurement of clinical progression in Huntington&#39;s disease patients.
Patients receiving the high dose of AMT-130 also showed near-baseline stability in motor and cognitive function throughout the 24 months of follow-up. Additionally, a statistically significant reduction of Neurofilament light chain (NfL)&nbsp;protein in cerebrospinal fluid (CSF) was observed in patients treated with AMT-130.
These patients had a mean reduction in CSF NfL of 11% compared with baseline at 24 months. Treatment with both dose strengths of AMT-130 resulted in mean CSF NfL levels below baseline at 24 months.
CSF NfL is an important biomarker of neurodegeneration and strongly associated with the clinical severity of Huntington&#39;s disease. An independent natural history study showed a 26% increase in CSF NfL at 24 months in patients with early manifest Huntington&#39;s disease.
Per the updated data, AMT-130 continues to be generally well-tolerated with a safety profile that is manageable at both doses. No new drug-related serious adverse events were reported.
We remind the investors that uniQure enjoys the FDA&#39;s Regenerative Medicine Advanced Therapy (RMAT) designation&nbsp;for AMT-130 to treat Huntington&#39;s disease.
The company is currently gearing up to hold an RMAT meeting with the FDA in the second half of 2024 to discuss these updated positive data seeking potential expedited clinical development pathways and accelerated approval for AMT-130.
In the second half of 2024, the company expects to complete enrolling patients in the third cohort of the&nbsp;U.S.&nbsp;phase I/II study exploring AMT-130 in combination with immunosuppression. Safety data from this cohort is anticipated in the first half of 2025.

UniQure Soars on New Huntington's Disease Study Data

Merck (NYSE: <a class="ticker" href="https://www.benzinga.com/stock/mrk#NYSE">MRK</a>) announced that the U.S. Centers for Disease Control and Prevention&#39;s (CDC) Advisory Committee on Immunization Practices (ACIP) has unanimously voted to recommend MRK&#39;s newly approved 21-valent&nbsp;pneumococcal conjugate vaccine (PCV) Capvaxive. The FDA approved Capvaxive earlier this month for the prevention of invasive pneumococcal disease and pneumococcal pneumonia in adults. Capvaxive became the first PCV specifically designed for adults to be approved by the FDA.
Capvaxive targets serotypes that account for approximately 84% of all invasive pneumococcal disease in older adults (50 years and older) in the United States, including eight serotypes not covered by currently licensed vaccines.
The ACIP recommends a single dose of Capvaxive for three distinct patient populations, the first being adults aged 65 years and older who have never received a pneumococcal conjugate vaccine or whose vaccination history is unknown. The second eligible patient population includes adults aged 19-64 years with certain underlying medical conditions or risk factors who have not previously received a pneumococcal conjugate vaccine or whose vaccination history is unknown.
The final patient population for whom ACIP recommends Merck&#39;s Capvaxive are adults aged 19 years and older who have begun their pneumococcal vaccine series with 13-valent PCV (PCV13) but have not received all the recommended pneumococcal 23-valent polysaccharide (PPSV23) vaccine doses.
Additionally, the ACIP advises that shared clinical decision-making should be applied for the use of a supplemental dose of Capvaxive for adults aged 65 years and older who have already completed their vaccine series with both PCV13 and PPSV23.
These new recommendations highlight Capvaxive&#39;s potential to enhance protection against pneumococcal disease in older adults and those with specific health conditions. However, these recommendations are provisional, which means they are not legally binding. These will only be official when the CDC reviews and finalizes the same.

Novo Nordisk acquired ocedurenone from KBP Biosciences in 2023. The phase III CLARION-CKD study, conducted by KBP Biosciences, aimed to assess ocedurenone&#39;s efficacy inmore than 600 enrolled patients with uncontrolled hypertension and advanced CKD, a condition for which effective treatments are critically needed. The study included a prespecified interim analysis after 12 weeks of treatment for all patients.
The decision to stop the trial underscores the challenges in developing effective therapies for complex conditions like advanced CKD and uncontrolled hypertension. The failure of ocedurenone brought into question the viability of the KBP Biosciences deal.

Novo Nordisk's Hypertension Drug Study Fails to Meet Goal

Intellia Therapeutics&#39; (NASDAQ: <a class="ticker" href="https://www.benzinga.com/stock/ntla#NASDAQ">NTLA</a>) shares gained 10% on Jun 3 after the company reported encouraging long-term data from the phase I portion of its early to mid-stage study evaluating NTLA-2002 for treating hereditary angioedema. Shares of NTLA continued to gain another 1.1% in the after-market hours.
NTLA-2002 is Intellia&#39;s investigational single-dose CRISPR/Cas9 gene-editing therapy, which is being developed as a one-time cure for HAE. It is a rare genetic condition that is marked by severe inflammatory attacks in various body organs and tissues, which can be painful and even life-threatening. Despite the current treatment options that require chronic administration for disease control, HAE attacks still occur.
NTLA-2002 aims to prevent HAE attacks by suppressing the plasma kallikrein activity. Per the management, the long-term data presented at a recent medical conference has bolstered the candidate&#39;s potential as a groundbreaking treatment for HAE as patients experienced durable elimination of their attacks in the ongoing phase I/II study.
Year to date, shares of Intellia have plunged 22.9% compared with the industry&#39;s 6.6% decline.

A total of 10 patients are being treated in the phase I portion of the study with either of the three one-time doses (25 mg, 50 mg, or 75 mg) of NTLA-2002,administered via intravenous infusion. Eight of these patients experienced no attacks during the initial 16-week observation period. These patients have experienced ongoing attack-free durations of greater than 18 months.
After the last assessment of HAE attacks as of&nbsp;Feb 12, 2024, a 98% mean reduction in monthly attack rate and a 99% mean reduction in moderate to severe attacks were observed after a single dose of NTLA-2002 in the overall patient population. Robust and sustainable reduction in HAE attack rate was observed upon treatment with all dose levels.&nbsp;
Intellia reported that the longest attack-free interval for an individual patient post-infusion is more than 26 months and ongoing. Treatment with NTLA-2002 has also shown a persistent reduction in HAE attacksin patients with the most severe HAE symptoms.
In the phase I/II study&#39;s beginning, two patients had the highest historic monthly HAE attack rates (16.8 and 14.0 attacks per month, respectively). Both patients were attack-free by the end of the 16-week primary observation period and have remained free of attacks through the latest follow-up.&nbsp;
Furthermore, Intellia reported that the longest attack-free duration among these two patients is 23.5 months and ongoing.&nbsp;All patients who discontinued prophylaxis treatment after NTLA-2002 remain free of chronic prophylaxis treatment.
At the time of the latest follow-up, the mean reduction in plasma kallikrein levels from baseline upon treatment with NTLA-2002 was 60% (25 mg, 88 weeks), 88% (50 mg, 72 weeks) and 95% (75 mg, 88 weeks).
Per Intellia, all three doses of the gene therapy candidate were well-tolerated in the phase I portion of the early to mid-stage study and most adverse events were mild in severity.
The company plans to assess HAE attacks and plasma kallikrein protein levelsthrough the remainder of the two-year follow-up period.
It has completed enrollment in the phase II portion of the study that will further evaluate the 25 mg and 50 mg doses of NTLA-2002 for HAE, expecting to report top-line data soon. Intellia also anticipates initiating a pivotal phase III study of the candidate for the same indication in the second half of 2024, subject to regulatory feedback.
Please note that the company&#39;s clinical-stage pipeline comprises another gene-editing candidate, NTLA-2001, which is currently being evaluated in a late-stage study in partnership with Regeneron to treat transthyretin amyloidosis with cardiomyopathy.
